Filling in walls

I know this question is probably beyond simple for most of you ,but I'm just a novice builder so here goes. 
I am going to attempt a small timber frame shed - 12x16. Two bents 12' wide. So 16' openings along the side. HAvent decided what the posts are going to rest on but am thinking cement slab. So whats the best way to board up the 16' spans. Stud up a wall as if regular stick building? Or can i put a post halfway between the corner posts. Then toenail some horizontal studs between them. Whats the conventional way of doing things?
