Can you tell by just looking at it?  I have a Ford 545D with the FEL running off the front pump that runs nothing else.  Would the difference be in the number of hoses attached to the valves?

It is just a port sometimes labeled pb or something similar. I have a Ford 340b which is older then your machine but it has a power beyond port on the loader valve that feeds the factory backhoe if I remember correctly.  They did put backhoes on the 545 so i would say it's possible if they continued to plumb them the same way.
I am not a hydraulic expert and could be wrong but if it is currently operating you would have 7 hoses coming out of the valve.  The power beyond may just return.  If it is a port that is just plugged I believe you need a "sleeve" to put in it to make it work as once it is operating you can't just plug it or it would dead head

"PB" or "BEY" for beyond is a definite.  Youll have an SAE plug fitting in a large port on an endcap.  It may require a sleeve threaded into the wall behind it to convert.

From memory the 545D was never offered with a backhoe. I also don't believe the loader valve has power beyond capabilities.

On that 545 with the front pump is it possible to feed an open center spool valve before it feeds the loader valve?

Again I'm no expert but if that first valve had a power beyond port you could use that to feed the loader valve. You still would need a return for that first valve as well. Surplus center sells a 3 spool loader valve.  2 are on a joystick the other a lever to run something else

What eric said.  As long as the lead valve has power beyond, a dedicated exhaust to tank line of its own, and has the highest pressure relief setting of the whole string.  Meaning you cant have an 1800 psi device plumbed upstream of a 2500.  The 1800 up front will limit the entire string of valves down the line to that pressure.
